__OPLC system with necessary hardware and some supplies for routine operation in sample separation.

This equipment performs a 2-dimensional separation technique compared to standard high performance liquid chromatography (HPLC), which is a 1-dimensional separation technique. HPLC is not a suitable option for the complexity of preparative work necessary for this project. In essence, OPLC combines all capabilities of HPLC, with those of thin layer chromatography (TLC) and Flash chromatography. As such, OPLC is the marriage of three techniques that allow unequaled preparative capabilities.____________________________________________________________

Suitable OPLC unit should be capable of using 5 x 20 cm, 10 x 20 cm and 20 x 20 cm size flat columns, and span a variety of stationary phases for normal and reverse phase operation, and include chiral operation potential. The unit should be pressurized to 50bars/725 psi and obtain this pressure in 45 s. The unit should allow preparative scale separations, depending on the column, as follows: 50 mg for 5 x 20 cm columns, 100 mg for 10 x 20 cm, and 200 mg for the 20 x 20 cm columns. A glass backed column should achieve separation of 500 mg. The nit should have unidirectional, 2-directional, 2-dimensional and circular separation capabilities. Solvent should be deliverable by HPLC pump. Sample insertion should be accomplished by either sample injection or direct sample application to the sorbent bed. OPLC should be capable of being interfaced with other laboratory detection instruments, such as mass spectrometers, UV/Vis and fluorescence spectrometers. Columns should be removable from the system and any stage during a separation.
